Tirzepatide Reconstitution Protocol
Tirzepatide is supplied as a lyophilized powder requiring reconstitution with bacteriostatic water before use in research applications. As a larger peptide (4,813.45 Da, 39 amino acids) with a C20 fatty diacid modification, tirzepatide has specific handling requirements that differ slightly from smaller peptides. This guide provides the complete reconstitution protocol for BeaCapra's 5 mg tirzepatide.
Equipment
- Tirzepatide lyophilized vial (BeaCapra 5 mg)
- Bacteriostatic water (0.9% benzyl alcohol)
- Sterile syringe (1-3 mL, Luer-lock)
- Sterile needle (25-30 gauge)
- Alcohol swabs (70% isopropyl)
- Nitrile gloves (powder-free)
- Clean, level work surface

|---|---|---|---|
| 1.0 mL | 5.0 mg/mL | 0.50 mg (500 mcg) | 0.25 mg (250 mcg) |
| 2.0 mL | 2.5 mg/mL | 0.25 mg (250 mcg) | 0.125 mg (125 mcg) |
| 2.5 mL | 2.0 mg/mL | 0.20 mg (200 mcg) | 0.10 mg (100 mcg) |
| 5.0 mL | 1.0 mg/mL | 0.10 mg (100 mcg) | 0.05 mg (50 mcg) |

Step-by-Step Protocol
1. Equilibrate
Remove the tirzepatide vial from -20°C storage and allow it to reach room temperature (15-25°C) for 10-15 minutes. Inspect the lyophilized cake — it should appear as a white to off-white powder or compact cake.
2. Prepare
Clean your work surface with 70% isopropyl alcohol. Put on nitrile gloves. Swab the rubber stoppers of both the tirzepatide and BAC water vials with alcohol wipes. Allow to air dry completely.
3. Draw Solvent
Using a sterile syringe and needle, draw the calculated volume of bacteriostatic water.
4. Add to Vial
Insert the needle through the tirzepatide vial's rubber stopper. Direct the stream against the glass wall — not onto the lyophilized powder. Depress the plunger slowly and steadily.
5. Dissolve
Remove the needle. Hold the vial at a slight angle and swirl gently in circular motions for 60-90 seconds. Do not shake, vortex, or agitate vigorously. Tirzepatide's C20 fatty acid chain is susceptible to aggregation at air-liquid interfaces created by bubbling.
If the powder does not fully dissolve, set the vial on a flat surface for 5-10 minutes, then swirl again. Complete dissolution may take slightly longer than smaller peptides due to tirzepatide's higher molecular weight.
6. Verify
The solution should be clear, colorless, and free of visible particulates. Discard if cloudiness or particles persist.
7. Label and Store
Label: compound name, concentration, reconstitution date, expiration (date + 28 days). Store at 2-8°C. Do not freeze.
Tirzepatide-Specific Handling Notes
Fatty Acid Sensitivity
Tirzepatide's C20 fatty diacid modification makes it slightly more susceptible to surface adsorption than non-acylated peptides. Low-bind tubes or glass vials are preferred for aliquoting. Avoid excessive transfers between containers, as each transfer loses a small amount of peptide to surface binding.
Solubility
Tirzepatide is freely soluble in aqueous solutions at standard research concentrations (0.5-5.0 mg/mL). No co-solvents are required. At very high concentrations (>10 mg/mL), viscosity may increase; this is normal for large acylated peptides.
Stability Window
Reconstituted tirzepatide in BAC water is stable for 28 days at 2-8°C — slightly shorter than some smaller peptides due to the compound's size and complexity. Mark your expiration date and plan accordingly.
Storage Summary
| Form | Temp | Duration |
|---|---|---|
| Lyophilized | -20°C | 24+ months |
| Lyophilized | 2-8°C | 6-12 months |
| Reconstituted (BAC) | 2-8°C | 28 days |
| Reconstituted (sterile H2O) | 2-8°C | 48 hours |
Frequently Asked Questions
How much water do I add to a 5 mg tirzepatide vial?
It depends on your desired concentration. Adding 2.5 mL of BAC water to a 5 mg vial produces a 2.0 mg/mL solution, where each 0.1 mL contains 200 mcg. Use our calculator for your specific needs.
Is tirzepatide reconstitution different from semaglutide?
The basic technique is the same (BAC water, swirl gently, don't shake), but tirzepatide may take slightly longer to dissolve due to its larger size (4,813 vs 4,114 Da). The reconstituted stability window is also slightly shorter (28 vs 30 days).
Can I freeze reconstituted tirzepatide?
No. Freeze-thaw cycles cause aggregation and reduce potency. Keep reconstituted solution at 2-8°C and use within 28 days. If your protocol runs longer, reconstitute a fresh vial.
What if the solution is cloudy after reconstitution?
Allow the vial to sit undisturbed at room temperature for 10-15 minutes, then swirl gently again. Persistent cloudiness or visible particles indicate potential degradation — do not use. This can happen if the powder was exposed to heat, moisture, or repeated freeze-thaw before reconstitution.
Can I use sterile water instead of BAC water?
Yes, but stability drops to approximately 48 hours without the benzyl alcohol preservative. BAC water is strongly recommended for multi-use vials.
