Mechanism of Action

Adipotide (CKGGRAKDC-GG-D(KLAKLAK)2) is a chimeric peptidomimetic with two functional domains linked by a glycine-glycine spacer.

Homing Domain (CKGGRAKDC): This cyclic peptide sequence selectively binds prohibitin, a protein highly expressed on the luminal surface of white adipose tissue vasculature endothelium but not on endothelium of other tissues.

Proapoptotic Domain (D(KLAKLAK)2): Once internalized via receptor-mediated endocytosis, this amphipathic D-amino acid peptide disrupts mitochondrial membranes, releasing cytochrome c and activating the intrinsic apoptotic cascade.

Vascular Remodeling: Endothelial cell apoptosis causes vascular regression in white fat depots, depriving adipocytes of blood supply and leading to adipocyte death and fat pad involution.
