Mechanism of Action
Melanotan I (Afamelanotide) is a synthetic tridecapeptide analog of α-MSH with an amino acid substitution (Nle4, D-Phe7) conferring enhanced potency and MC1R selectivity.
MC1R Activation: MT-I binds MC1R on melanocytes, activating Gs-coupled adenylyl cyclase. Increased cAMP activates PKA, which phosphorylates CREB, upregulating MITF and downstream melanogenic enzymes (tyrosinase, TRP-1, TRP-2).
Eumelanin Synthesis: MC1R activation shifts melanin synthesis from pheomelanin (red/yellow, photo-damaging) toward eumelanin (brown/black, photoprotective), increasing constitutive UV protection.
Enhanced Stability: The Nle4 substitution prevents methionine oxidation and D-Phe7 increases proteolytic resistance compared to native α-MSH, extending biological half-life.
