Research Benefits
- Neuromuscular Junction Research — SNAP-8 (Acetyl Octapeptide-3) modulates SNARE complex assembly, providing a topical research tool for studying neuromuscular signal transduction.
- Anti-Wrinkle Mechanism Studies — By competing with SNAP-25 for SNARE complex formation, SNAP-8 reduces vesicle fusion and neurotransmitter release at the dermal neuromuscular junction.
- Cosmeceutical Research — SNAP-8 is extensively studied as a topical peptide for reducing expression lines, with published studies demonstrating measurable effects on skin surface topology.
- Non-Invasive Alternative — As a topically applied peptide, SNAP-8 provides a non-invasive model for studying SNARE-mediated exocytosis modulation.
Mechanism of Action
SNAP-8 (Acetyl Octapeptide-3) is a synthetic peptide that mimics the N-terminal domain of SNAP-25, a key component of the SNARE (Soluble N-ethylmaleimide-sensitive factor Attachment protein REceptor) complex.
SNARE Complex Competition: SNAP-8 competes with native SNAP-25 for binding positions in the SNARE complex, which consists of SNAP-25, syntaxin-1, and VAMP-2 (synaptobrevin). By partially displacing SNAP-25, it destabilizes the complex and reduces vesicle-membrane fusion efficiency.
Reduced Exocytosis: Destabilized SNARE complexes result in diminished acetylcholine release at neuromuscular junctions in the skin, reducing muscle contraction intensity and associated expression lines.
Topical Penetration: The acetylated N-terminus enhances lipophilicity, improving stratum corneum penetration. The eight-amino-acid sequence is small enough for transdermal delivery to the dermal-epidermal junction.
Comparison to Botulinum Toxin: While botulinum toxin cleaves SNARE proteins irreversibly, SNAP-8 acts as a competitive inhibitor with reversible, dose-dependent effects on vesicle fusion.
Research Protocol
Derived from published research. For research use only.
Preparation
SNAP-8 is typically dissolved in aqueous formulations at 3-10% w/v for topical application studies. It is water-soluble and does not require organic solvents.
Research Applications (from published studies)
- Topical studies: 3-10% SNAP-8 in cream or serum base, applied twice daily for 28-56 days with silicone replica or profilometry assessment.
- In vitro neuromuscular: 50-500 μM in primary neuron/myocyte co-cultures, measuring ACh release via patch-clamp or calcium imaging.
- Skin penetration: Franz diffusion cell with dermatomed skin, 5% SNAP-8 donor solution.
Storage
Store lyophilized at -20°C. Solutions at 2-8°C are stable for 30+ days.
Safety & Handling
Research Use Only.
Storage Conditions
- Lyophilized: -20°C
- Aqueous solutions: 2-8°C, stable for 30+ days
- Compatible with most cosmetic formulation excipients
Known Considerations from Literature
- SNAP-8 is designed for topical application. Systemic administration is not typical in published research.
- No significant skin irritation or sensitization has been reported in published dermal tolerance studies.
- Effects are reversible and dose-dependent, unlike irreversible botulinum toxin cleavage.
Handling
Standard PPE. No special ventilation required for aqueous solutions.
Frequently Asked Questions
- What is SNAP-8?
- SNAP-8 (Acetyl Octapeptide-3) is a synthetic peptide that mimics part of the SNAP-25 protein to competitively modulate SNARE complex assembly, reducing neurotransmitter-mediated muscle contraction at the skin level.
- How does SNAP-8 differ from botulinum toxin in research?
- Botulinum toxin irreversibly cleaves SNARE proteins (specifically SNAP-25 or VAMP-2 depending on serotype). SNAP-8 is a competitive inhibitor with reversible effects, does not require injection, and has no paralytic activity.
- Is SNAP-8 water-soluble?
- Yes. SNAP-8 is fully water-soluble and compatible with aqueous formulations, making it suitable for topical research preparations without organic solvents.
- What purity is this product?
- BeaCapra SNAP-8 is ≥99% pure by HPLC with COA documentation.
Customer Reviews
Exceptional purity and fast shipping
Third-party COA verified at 99.1% purity. Arrived in 2 days with proper cold packaging. The documentation included was thorough and professional. Will be ordering again for our lab.
Best supplier I've found
After trying several suppliers, BeaCapra consistently delivers the highest quality compounds. The subscribe & save option is a great value, and their customer support team answered all my questions promptly.
Great product, COA checks out
Ran our own HPLC analysis and confirmed the purity claims. Packaging was secure and discreet. Only minor note — would love to see more size options for some of the newer compounds. Otherwise excellent.
Subscribe & save is the way to go
The 15% discount on subscription makes this the most competitive pricing I've found for research-grade compounds at this purity level. Auto-delivery every 30 days has been seamless. Highly recommend.
Exactly what our research team needed
We've been sourcing from BeaCapra for our university lab and the consistency batch-to-batch is impressive. Every order comes with a COA and the compounds are properly lyophilized. Professional operation all around.
